Be sure to have fun and don't try to do too much on your first day. Since you have never been before, you may want to ask someone to show you around a bit and teach you how to use some of the equipment. Once you find something you like, stick with it. That is the important part.

My wife and I were both semi-elite athletes at one point, but our work schedules and children do not permit us to exercise like we did when we were younger (i.e., 2+ hours per day). We both put on some weight during my wife's first pregnancy, and getting it off has been a struggle. Nonetheless, we are both back to our pre-children weights, and we have both started early morning exercise routines to get back into shape. While it is unlikely that we will ever be as fit as we were when we were younger, I have seen some pretty marked improvement in both appearance and fitness even in the short amount of time we have been exercising seriously.
The important parts - for me at least - have been: (1) not mentally punishing myself for missing a work out; (2) not trying to puch myself the way I did when I was younger; and (3) diversifying my workout routine. So far, following these simple rules has helped me fit in four or five 20 to 30 minute workouts per week for the past few months (which, while not as much as much exercise as I would like, is a lot better than nothing at all).
