I thought this might be an interesting thread/discussion. What are some of the most memorable "OH SHIT" moments of gaming for you so far? Here are a couple of mine:
Final Fantasy (NES) - The class upgrade. This honestly blew me away.
Mega Man X3 - Being able to play as Zero for the first time. Truly awesome.
Mega Man 9 - Being able to play as PROTO MAN. Fuck yes.
Xenogears - Any time Grahf shows up/Grahf's theme plays.
Contra: Hard Corps - The first level in general.
Metal Gear 2 (MSX) - The final boss fight against Big Boss. Anyone that's played the game will know exactly what I mean.

Mario 64.
One of the first 3D games that had a HUGE mainstream success I believe.
I didn't even own an N64 but I watched my next door neighbors play it in awe of everything about it.
I remember one time playing some demo that was set-up at the local mall for it's release and some old woman coming up behind me going: "Woooow! Can you make that little man jump higher? Make him jump over there! What's his name!?"
I never saw an old person so engrossed in a video game. I was about to let her play - but then again I was also kind of annoyed by her. I just remember everyone being completely impressed with that game and the N64 because of it.
Of course Sony came along and stole much of it's thunder after that - but...
EDIT: Sorry if this isn't what you meant by video game "moments" It's more of a moment in time I guess.

That was sort of cool because my brother's girlfriend at the time bought that game for him and he smoked. He had also gotten the strategy guide for it and so it smelled like cigarettes. Usually that's a bad thing but I felt it was rather appropriate.

That time when you play Crazy Taxi for the first time, and then like, every other time you ever play Crazy taxi in your whole life.

In Bioshock it was when I accidentally pissed off the first neutral big daddy. I hadn't expected him to not be hostile. I figured he didn't know I was there and would get the drop on him. That ended poorly.
On ones first play through of Demon's Souls it's every fifteen minutes of gameplay.
In Megaman X it was after discovering I could get the Max Megabuster BEFORE Zero gets beat up.
In Syphon Filter it was holding the taser too long so that the enemy lit on fire. I had to put the controller down and stop playing for a moment. It felt like I had committed a crime. For some reason that was just so much worse then shooting guys in the face.
Too many to even name.

Great topic, but damn I can't think of much at the moment but I'm sure a lot of stuff will come to mind eventually.
If this fits though, for something that's one of the most frightening, weirdest, craziest wtf subtle moments in gaming history for me... how about the dead guy in front of Kraid's door in Super Metroid? Who or what was he? Why was he there? What happened?!?!? Ah the wondrous subtle moments of Super Metroid.
- Deus Ex ... Deadalus, Icarus, and Helios. Nuff' said.
- System Shock 2, too many moments to count and it's just best to experience this beast if you haven't yourself.
- Unreal. Finally played it last year, running around the interior of your crashed ship at the beginning thinking "hey this is pretty neat", then suddenly ... you step outside of the ship with that music kicking in, looking around the massive alien environment around you with multiple moons and stars in the skies, you realize YOU have just been teleported to another world and are in for one insane ride. The games storyline was about as simplistic as Doom or other old sidescrollers, but the atmosphere and the little details here and there were so powerful it was an experience that let you interpret things how you wanted with your own imagination. Damn do I miss games that could do this.

When the PS3 vs Xbox 360 argument launched and one of the biggest claims was that PS3 was going to have FFXIII exclusively and then it was announced for both systems. Made me
hard.

The climax of act 3 of Dragon Age 2. If you've played it you know what I'm talking about.
