I thought this would be fun and light topic , I always thought of it.
We all know that a lot of games succeeded(financially) based on luck , like nintendogs and Carnival games given there are similar games,Why those are the popular one? God knows, just like how god knows why facebook and myspace are the top social networking sites given others do exist. On the opposite, there are great games that simply do not sell for reason we do not understand like psychonauts and beyond good and evil.
So you are now head of Ubisoft(or EA) , and the company is in financial trouble , and all the investors and fans of your company are depending on you to save their favorite company and their investment. You have $40 million budget, which you don't have to use all of it, to create the next blockbuster top selling game to bring the company back to its feet.
What game will you create? What genre? What is the setting? Which platforms will you choose to release the game on? What are the most important feature of the game? Why do you think this game will be the next "Halo, Super Mario, MW, GTA, WoW" ?
Its fun to see the different perspectives of different people, and remember this is for fun so you can't go wrong.

The game I would make would be a FPS, set in a future that allows for space ships, aliens and space colonization, but near enough to the present to allow for the weapons to be familiar as well as other things - such as brand sponsors.
The game will consist of a war between Space Pirates, Government Sponsered Assassins (We'll call them "Space Ninjas"), and between the fighting, a rare mutagenic virus breaks out that starts infecting many and drives them insane (We'll call the "Space Zombies").
Despite the huge clash going on, we won't make this a humorous take. Oh no, this will be serious. Extremely serious. Grim and dark, with no colors other than black, gray and brown.
However, none of this is really going to matter, as we'll only make the single-player mode about 5 hours long. All the money will really be going into the multiplayer mode, which will only have 5 unique maps upon release. However, we'll make them the best we can, and after less than one week of the game being out, we'll release another 5 maps that cost $10. We'll continue to make more DLC post release, containing everything from player skins to new weapons.
Moreover, every game will ship with a unique code that, when entered, will give you instant access to enough Experience that will make you an unstoppable killing machine. Also, those who enter the code will instantly get a free 100G Achievement/Gold Trophy, that will only be obtainable to the rest after the game has been out for two years - meaning the only way to get this stuff is to buy it new.

I've thought of this same topic for years now.
After being disappointed with Kayne and Lynch I thought of this idea.
You are a bank robber, not in the wild west but in modern time, and you go after big heists. As a bank robber you have to case the bank you want to rob, you will watch traffic patterns in and out of the building, you will watch employee habits (when they clock in, when they clock out, when they go on break, when the bank has the most employees on staff, etc), you will have to acquire a schematic of the bank's security system, as well as, a copy of the bank's building layout (blueprints).
After casing the place you decided what is needed to finish the job. You may need to higher other guys that know how to get around or disable high end security systems, or the best route could be to higher a guy to get a job at the bank, build some rapport with the company and have them let you in the building after closing. If the building is big enough you may have to higher a group of thugs to help you hold the place up. The guys you higher woud be completely up to you, there wont be any right or wrong way to rob the place but some ways may be easier than others.
As you rob more and more buildings, you get tipped off to better jobs and as your reputation builds your access to better criminals for higher gets broader.
The game would play out more like a puzzle/strategy game than an action title. I think the game could be fun, if the right team were to handle it. I was thinking Ubisoft, they seem to be really good and making obscure games, or trying new formulas out.

For a few years I thought about making a 16-bit RPG (aka SNES-era Final Fantasy ripoff) that was based on the Friday The 13th series.

Since I want to make money in this scenario, and not necessarily the best game, I would look around at what is popular and steal everything. So, basically my game would have zombies, wizards, space marines, nazis, and big tittied women that street fight. Also, the cover would have John Madden in the corner.

I'd make a handful of casual puzzle games. Then use the remaining money and profits to make the most expensive and ambitious sprite based game ever.

Largescale FPS for lanparties. Think Half-Life meets Planetside, something that can accommodate 500+ players on a dedicated server. Maps based on real life cities. Really it would be something like an FPS version of the book DMZ or a large scale version of Battlefield 2142 but it would play alot smoother.

I would make a game like fallout 3 except without a such a freakin low level cap!
Now, not to be offensive in anyway shape or form, but if you take a look at the movie Pan's Labyrinth you'll notice that it's overwhelming success was linked to not how good of a movie it was (because it was a decent movie at best imo), but in it's direct appeal to spanish speaking people.
I don't have the details, but i would push for an action rpg of sorts that would appeal directly to the culture and language of the spanish speaking people of the world.
