I agree with dsheinem that we may need to cap these kind of threads, perhaps someone can pin-up a dedicated thread for getting in with each systems (a 101 guide without any fanboyism).

Back to the OP, which version of PS3 (specifically if its B/C with PS2 games), and how many storage space you got? These two can factor a lot.

And always, which genre appeals to you the most? There are plenty of great games in each genre for the PS3, thou I would say the JRPG library is much smaller than the PS2.

Uncharted (the first 2 at least) and the God of War's have already been mentioned but I'd reiterate to try those for sure. Some others I would recommend:

-Dead Space (at least the first one)
-Borderlands (I was late to this series, so I played the 2nd one before the 1st but both are great games. The 2nd one is superior IMO though)
-Darksiders
-Batman: Arkham City

Those are the ones off the top of my head. Lots of great games.

Games/series that I would recommend checking out. Don't really have a ton of PS3 exclusives in my collection but here are a few games to check out.

1. Uncharted
2. Infamous
3. God of War
4. Heavy Rain(Altough only if you don't mind QTEs and the most boring tutorial level ever)


Also I just picked up a couple of PS3 games on the cheap if your interested. Got copies of Yakuza 3, Valkyria Chronicles, Star Ocean the Last Hope International, Heavy Rain, ES Oblivion GOTY, Skyrim, and Fallout 3 GOTY.
