I was thinking about purchasing Elder Scrolls V Skyrim because I am interested in the mods that the hacker community has accomplished. Many of them require to have the latest STEAM patch.
I was wondering, if you want these types of mods, do you have to purchase it directly from the STEAM website or can you buy the game at the store or online in which you can sign up?
This game looks really fun, I'm curious about trying this along with Dragon's Dogma.

When you install a retail copy it will automatically associate itself with your Steam account and you would be able to snag mods for it no problem.

So I would be able to purchase the game from eBay or go to Wal Mart and buy it it would give me the option to sign up to STEAM? Thanks, I didn't know that Steam was involved with Skyrim. I was told that you have to buy the $60 pack online because there was some signing up or whatever. Oh, one more thing, this also includes used Skyrim games correct? or do I have to purchase a new one because of key issues?

There's no such thing as a used copy of Skyrim for PC.
It's a Steamworks game (like these). You can buy a retail copy, but as part of the installation process you bind the included serial number to your Steam account. Until such time as you can unbind a copy of a game from a Steam account and resell it (if ever), that means new copies only.
Once it's bound to your account, it's really no different from a game simply purchased on Steam. It's noted in the Licenses + Subscriptions part of your Account page, but so are gifts, or free things. You can redownload the game without the disc, it still updates the same, all that.
Basically, it's a retail Steam code, accompanied by an installer cache on a disc, and whatever packaging/printed materials the game has. If you don't care about that, you might as well buy the game on Steam itself if it's priced well.
Personally, particularly if I'm paying full price, I tend to buy Steamworks games at retail. Just 'cause. Well, Skyrim came with a fairly nice map for preorders/first run, there was that

It's really awesome on Steam because of the Steamworks integration. It makes adding mods super simple. I love it.

IN CONCLUSION:
I bought the game and I can say with having all those different mods into it, it's like playing a completely different game. When I played Diablo II I loved playing the druid because having a werewolf warrior was the funnest thing imaginable. Skyrim adds that dimension to a whole new level!
Excellent game, I still need to finish it, but before I do I need to open up a post about streaming HDMI from my PlayStation 3 to my PC because I want to do some youtube vids =]
