Good question...
I'd say:
NES: DoubleDragon (1 or 2), Mario 2, 3, Batman
General: SuperStreetFighter 2 turbo, PacMan, GTA SanAndreas(or the most recent with lots of AI and NPC AI)

Hmm a number of games are coming to mind Super Street Fighter 2 Turbo , Street Fighter Alpha 2 ,Soul Calibur 2, Tekken 3 , Tekken 5 , Tekken 6 but if I had to choose just one I guess it would be Super Street Fighter 2 Turbo as SSF2T had universal appeal all of my friends and family could play it but the others like Alpha 2 and Soul Calibur were limited to consoles and the Arcade players or my friends didn't really get into them games in fact they near enough stopped playing games when Arcades went out.
So yeah if it had to be just ONE Super Street Fighter 2 Turbo

Starfox 64. I've beaten the game something like 5 dozen times(pretty easy given its only an hour long), and it only gets better with every play through. It is pretty much my idea of a perfect game. Well, except for that Aquas level, that one was kinda meh.
Current systems: Atari 2600[x2], NES(top loader), SNES, N64, Gamecube, Wii, Saturn, Dreamcast, Playstation, PS2, PS2 slim, xbox, xbox 360, Turbografx-16, Game Boy Pocket, Game boy SP[x2], Nintendo DS[x2], Nintendo DSi, Nintendo 3DS, Game Gear, Neo Geo Pocket Color, Wonderswan Color, Neo Geo MVS.

If we are allowed Anthologies, then I'd pick the Metal Slug one. There's a lot of gaming in one of them, and collecting all the items as well as perfecting the levels would take plenty of time up.
I'd be tempted to agree with the Football Manager type of game though, they tend to be endless and highly addictive.

If I had an answer, it'd probably be Mike Tysons Punch-Out! Such a damn good game, and I was never a boxing fan.

I might have to go with Morrowind if I truly only had one game to play for the rest of my life. There is just so much unique content to see and do that it could take you years to do it. Not to mention all the characters and classes and specialties you could mix and match. You can even play the game in a non standard ways, like trying to kill every NPC in the game world.

Star Fox 64 and Mike Tyson's Punch Out are both good choices. It doesn't seem to matter how many times I beat those games, I still enjoy them.

Command & Conquer: Red Alert. With 2 campaigns, 1 short secret campaign, 100+ multiplayer maps that don't require an Internet connection, and around 50 stand-alone missions, the replayability is just insane. And that's why I'm playing it to this day, I actually played this afternoon.
Black DSi, Pink GBC, Softmodded Wii, Softmodded Fat Ps2, Black Gamecube, Xbox, Dreamcast, Ps1 Slim, Atomic Purple GBC, Onyx GBA SP, Gears of War 3 Edition Xbox 360 Slim, PSP 3000, NES.
