Castlevania Order of Ecclesia. If you even remotely enjoy the Castlevania series, you need to get this game.

Star Fox Command is pretty good in my opinion. It took a lot of ideas from Star Fox 2. You start off on a map and have to beat all the enemies. The battles are in all range mode.

Metroid Prime Hunters: Not the best Metroid game, but it has the best multiplayer shooting than any other DS game.

Mario Kart DS: One of the best games on the DS with great multiplayer, however playing online can get real shitty real quick.

Pokemon Platinum: No need for explaining why.

Warioware Touched: Great game, but avoid playing it in public.

Tony Hawks Downhill Jam: This was actually not half bad. If you liked the older Tony Hawk games, then you'll like this one too. Probably the best Tony Hawk game you will ever get on a mobile platform. Decent multiplayer, but you won't ever find anyone with another copy.

Nanostray I and II: Both are awesome. One of the few good shumps on the DS.
Systems: SNES, PS1, GBA SP, Gamecube, DS Lite, Wii, PS3 (Fully Backwards Compatible), Dreamcast, Nintendo 64, Sega Genesis, a Sega CD, and a Sega Saturn.

New Super Mario Bros. is definitely the best DS game. It truly brings mario back to his roots with 2D action and modern graphical flare.

I'm surprised no one has mentioned Mario VS Donkey Kong 2: March of the minis. This fun puzzle game plays a lot like lemmings. It's much better than the first Mario VS Donkey Kong for GBA. There's also a cool function that lets you build your own levels.

Viewtiful Joe - Double Trouble! is a great game if you like brawlers.

The World Ends With You is different take on RPGs and Squareenix really did a good job in this one.

The Legendary Starfy is a good 2D platformer with a starfish as the lead character.

Sonic Rush is a great 2D Sonic believe it or not.

Moon is a pretty good FPS that has some killer graphics considering or DSs limitations.

Lunar Knights is a Hideo Kojima game nough said.

Kirby - Canvas Curse is the reason I bought a DS. Shame it never had a sequel.

Dementium The Ward is a good horror title that never got any attention.

Brain Age - Train Your Brain in Minutes a Day! is great a must play.

These are just a few I thought of that were pretty good that are lesser known games.

I love Ketsui Death Label, but I know a lot of people who don't. All the DS Castlevania games are also highly recommended.

The wife was addicted to both Puzzle Quest games, as well as Yoshi's Island DS. She enjoyed the Professor Layton games, but ripped through them too quickly.

My favorites have been:

the DS Castlevania games (Dawn of Sorrow, Portrait of Ruin, Order of Ecclesia)
the Phoenix Wright games (Justice for All was rather forgettable though)
Advance Wars: Days of Ruin (Dark Conflict if you're in Europe)
The World Ends With You (despite needing two brains and four eyes to play properly)
Final Fantasy Tactics Advance 2
the Professor Layton games
Hotel Dusk room 215
Devil Survivor
