For some reason, I can only really think of three off the top of my head, although I'm sure there are some more:

1) Second world in Final Fantasy III.

2) Gran Pulse in FF XIII (I pity anyone who stopped playing before that).

3) Stepping off the boat in Shenmue II

Just one that I'm anticipating: Stepping out of the cave in Shenmue III.

It sticks out to me too, because it's easily the pinnacle of the game. Everything up to that point feels like it's really building toward something, and everything after feels like arbitrary, drawn out, fluff.

All new games have that moment anymore.

You know... when that day one 20GB update finally gets done and there's that incredible feeling you get when you can actually play something.
