I snagged a Zelda Amiibo to have an Amiibo to get to 50 and fight against. Can someone explain why people are trying to own them all? Is there a reasoning beyond the collection aspect?

From what I can gather, no. I think it's almost purely based on desire/rarity and "collection" purposes. Though actually i'm quiet surprised how great the figures look for their price and so they make for pretty good display. Personally, I'll own them for the bonus content in games such as Hyrule Warriors, Smash, and MK8, but other than that it's just for owning the figures themselves, displaying them, etc. I don't think i'll own them ALL, but just my favorites on top of any exclusives I can get my hands on. I'm a bit baffled by how much money some people are spending on the hard to find ones.

I am a bit excited to see what direction Nintendo takes Amiibo. I think it's proven to be a big financial success. I just hope it doesn't come down to them holding back content in their games to pressure people to buy more Amiibos in order to unlock "everything". Nintendos entrance into the DLC realm has been interesting.

I think a lot of people are grabbing them with anticipation that there will be an amiibo specific game. Add to it the difficulty in acquiring certain figures makes people a bit more desperate to catch them all. I have a co-worker who bought their first amiibo and then immediately cruised numerous stores to find more. He wasn't interested at all, but there is a gotta have it mentality that comes with them.

So what's the best way to open these things up, while still preserving the packaging as much as possible?

I open mine at the bottom. If you use an Xacto knife or something like that, cut through the bottom such that the perforated edge is a hinge, it works pretty well. Makes it pretty easy to take them out and put them back.

My largest concern with these things is dust accumulation. I don't want to have to take alcohol to them later or something.

Mine are kept in a temperature controlled negative air flow chamber - so no fears of dust.

Nice. Is there a trick to getting them to ship internationally? I have an Amazon jp account and have a saved address, but it's not wanting to accept it.

Right; you do need a proxy, shipping will be free to your proxy and then whatever SAL is to you ($6-7 for one and about $15 for 3). If you don't have a buddy in JP, you could always try to enlist Rancor - but $16 shipped for Rosalina is pretty solid.
