Usually when boredom sets in, which is around an hour or two, depending how into the game I am.
Skyrim's a good example - I must've put in over a hundred hours when I first played it (On Xbox 360), and did so again when I got it on PS3. After completing the main storyline and DLC, I grew bored with it and sending up selling it on, though I kept the save file if I ever grew the desire to ride dragons and shout at people in towns.

I personally like to try and finish every single game i play, even atrocities like Sonic '06 (sadly, didn't make it on that one.

I usually quit when i get to the point of thinking of other games while i play it.
Than, it's about time i stop.
I think, since 2013, I've beaten about 80 or so games. I've played around 120 though, so I beat more than I play.

Just assume you could be enjoying it a lot more once you actually FEEL like playing it, and hold it off, even if it means forever. Always rings true for me... games I come back to after even months after a time feel fresh and rewarding again versus when I left off. That extra little bit you need to do to finish off the game is probably the perfect sample size as well before it overstays it's welcome, versus starting over from the beginning.
With RPGs and those type of genres though, it can be kind of hard.
