No. His victims are never in on it-- that's the whole point of Bruno and his previous works, Borat and Da Ali G show. I know that in Borat, Pamela Anderson was in on the joke. If she hadn't been, Sacha Baron Cohen probably would have been shot.
Some people were actors-- like the boyfriend, the Mexican workers, and his assistant Lutz, but mostly only the people who need to be in on the joke are. After Borat came out, several people sued because they thought that the releases they signed were misleading (and they probably were), but I think they lost. I'm sure the same thing happened with Bruno.
