Mario and Luigi games have some very funny moments. Otherwise go for disgaea or wait for the newly announced Mugen Souls(traditional RPG with Disgaea level craziness).

The funniest I have played is Barkley: Shut Up & Jam Gaiden. I laughed the whole way through and the gameplay is surprisingly good too. Plus, it's totally free, so there is no reason not to play it:

Contact for the DS has a lot of humor to it, primarily in the vein of internet/gaming culture references.

Maybe it's just me... but I found the DS remakes of the Dragon Quest games to be pretty funny. Lots of subtle humor, I especially enjoyed the insults. I laughed so hard when somebody called me a "bog trotter".

The Penny Arcade games, if you're looking for something digital. I seem to recall the Shadow Hearts games having some humor to them, though they could get pretty dark too. We mentioned Disgaea, but pretty much all Nippon Ichi titles have a lot of humor. What I always liked about those games is characters who are well aware that they are in a video game, or at the very least aware that they are characters in a story. I loved when a certain powerful character in Disgaea 2 gets bumped down to level 1 and becomes furious at having lost all of her levels

I would also recommend any of the Working Designs games (especially the Lunar series) and Mario and Luigi.

Yeah, KoL was good fun for a bit. I played it for a couple months. I liked that you only got a certain number of turns per day kept me interested longer, but I eventually got bored with it.
