I'd recommend most of what's already mentioned here (Trauma Center, Phoenix Wright, Tetris DS, etc.) But a warning - the first Trauma Center game (Under the Knife) is CRAZY HARD once you get through the first couple of operations, and there's no way to change the difficulty. But because of this, you're guaranteed to spend at least 12 hours on the game just repeating certain operations over and over again until you pass them (or until you break your DS touchscreen in frustration... :P ) Trauma Center: Under the Knife 2 is easier, though.

You could also try Mega Man ZX and Mega Man ZX Advent. Nice graphics and music, nice length (it took around 12 hours for me to beat the first game) and they're not frustratingly hard.

I don't think New Super Mario Bros will last 12 hours. It's an awesome game, but my only gripe is the lack of a "new quest."

Get little Red Riding Hood's Zombie Barbeque it's fun as hell, cheap ($10 new at GS), has zombies in it and was reviewed by Classic Game Room.

Here's the link to GS's page on it:

And here's CGR's review:

The game is best described as a SHMUP meets House Of The Dead meets Robotron with zany bosses and awesome over the top action.
My Consoles:
Genesis - Nomad - SegaCD - GameGear - Sega Saturn - Dreamcast - NES - SNES - N64 - Gamecube - Wii - Playstation - PSone & LCD - PS2 - PS3 - Xbox - 3DS

That's the power of a busty Little Red Riding Hood, Zombies and the incredible Monotone mind control of CGR at work for you!

I watched CGR's review of the game awhile ago. Looks really fun. Might get it after X-Mas is over.

I'd avoid anything that's been recommended that has heavy touch screen emphasis. The lost precision in a car drives me insane. Also, I once made a pretty big scratch in my DS screen because of some idiot braking late on the freeway :( .

Love many of the recommendations thus far, especially Little Red (though not for the car), Phoenix Wright, Mega Man ZX, and Castlevania, but I've got to add Retro Game Challenge and Tokyo Beat Down  .

I would not play Trauma Center on a car ride (and I S-ranked all operations, including the extras after the story ended).

I don't think Kirby can last 12 hours - but it can if you replay it with all the unlocked characters (just the same levels again though).

Castlevanias though you certainly can. I have only played Dawn of Sorrow on the DS, but with the "soul collecting" and unlocked Julius mode it can last a good bit. You can also then do the Boss rushes and so on. I'm guessing the other DS Castlevanias are also good choices, but I haven't played them.
