I am having no problems. Bright rooms, dark rooms, extended period of time...no problems. The longest I played in one sitting was about an hour, and afterward I didn't feel any different from playing a regular DS. My fiancee doesn't have any problems either. My sister tried it for about 30 seconds and couldn't look at it anymore. She had to turn off the 3D.

You just have to find the setting right for you. Mine is maybe 1/3 of the way up, but it's just fine for me.

The lack of launch games was the biggest reason I didn't buy one. The other reasons being that I'll just wait for the next version of the 3DS to come out, and that I'd like to wait and see if a region-unlock hack comes out. JUS and JSS were the only games I really played on the DS anyway, unless we count the NES and Neo Geo emulators.
Anyone who does get a 3DS would do well to get SSF4. I'm not a fan of SSF4, but it would just seem silly not to get it if you are getting a 3DS at launch, as it seems head-to-toe meatier in terms of gameplay, replay and content.

Street Fighter gave me no problems when I played it for about half an hour, however Pilotwings hurt my eyes after just a few minutes.

I wasn't originally going to get Street Fighter, but my Amazon preorder credits made it $5, so I said might as well.

I'm having trouble with the 3D on Pilot wings, especially on the free flight mode. I think the thing that makes it near unbearable is the fact the you are trying to look at the goal in the distance, all the while your plane is jumping out of the screen in the foreground. I overcame the issue by either jumping to first person mode (plane only I believe) or by just turning the 3D off :S
Anyone else have fun with stresspass? I only found 2 people in my daily commute
Also, side question - what did you do with your old hardware? I'm considering selling my DSi, but couldn't give it to gamestop for only $75 :S

Don't get rid of your DSi if you plan to do any serious DS gaming. The compatibility mode sucks hard. You either get blurry upconversion, or very tiny 1:1 mode, that takes up about 1/3 of the screen only.
I found that with Pilotwings I have to use a lesser 3D value and hold it a little further away.

I played Pilotwings and Lego Star Wars for about an hour earlier today, both were pretty incredible. After the first time playing it I felt alittle uneasy (like I had a drink in me), but other than that it was fine. Ironically my eyes started to hurt in 2d mode, but not 3d. So I am cross as to why.
What launch titles are multiplayer? I know Tom Clancy is hotseat and Streetfighter is multicart.
