Yes, the rewrite is easily possible with the now-common protocol, but that is not afaik implemented through the GameCube cables. I think it might require voltages not possible through the controller port of a GameCube, though I'm not that knowledgeable on these things.

So a 5v EPROM will need 12v to be programmed. But Modern flash memories, 3.3v or 5v, use the same voltage for read and write operations. You just need access to specific pins.
